چکیده

Methods: Medial meniscectomy was used to induce OA in male Lewis rats. Therapy with zoledronate and human PTH was initiated immediately after surgery. A dynamic weight-bearing system was deployed to evaluate the weight-bearing capacity. At the end of the 10-week study cartilage pathology was evaluated by contrast μCT imaging and histology. Cortical bone geometry at femoral mid-diaphysis was evaluated by μCT and strength was tested using 3-point bending method.

Effect of antiresorptive and anabolic bone therapy on development of osteoarthritis in a posttraumatic rat model of OA

INTRODUCTION Osteoarthritis (OA) is a leading cause of disability, but despite the high unmet clinical need and extensive research seeking dependable therapeutic interventions, no proven disease-modifying treatment for OA is currently available. Due to the close interaction and interplay between the articular cartilage and the subchondral bone plate, it has been hypothesized that antiresorptive...

OPG and PTH-(1-34) have additive effects on bone density and mechanical strength in osteopenic ovariectomized rats.

PTH is a potent bone anabolic factor, and its combination with antiresorptive agents has been proposed as a therapy for osteoporosis. We tested the effects of PTH, alone and in combination with the novel antiresorptive agent OPG, in a rat model of severe osteopenia. Sprague Dawley rats were sham-operated or ovariectomized at 3 months of age. Rats were untreated for 15 months, at which time ovar...

Disuse in adult male rats attenuates the bone anabolic response to a therapeutic dose of parathyroid hormone.

Intermittent treatment with parathyroid hormone (PTH) increases bone formation and prevents bone loss in hindlimb-unloaded (HLU) rats. However, the mechanisms of action of PTH are incompletely known. To explore possible interactions between weight bearing and PTH, we treated 6-mo-old weight-bearing and HLU rats with a human therapeutic dose (1 microg.kg(-1).day(-1)) of human PTH(1-34) (hPTH). C...

Abaloparatide: a new anabolic therapy on the horizon.

Teriparatide administered by daily subcutaneous injection, was approved in the United States in 2002, and has been the only anabolic medication available in the United States for almost 13 years. Outside the US, teriparatide is joined only by intact PTH (1–84) as anabolic therapy for osteoporosis treatment. Although both anabolic and antiresorptive medications improve bone density and strength,...

Histomorphometric, physical, and mechanical effects of spaceflight and insulin-like growth factor-I on rat long bones.

Previous experiments have shown that skeletal unloading resulting from exposure to microgravity induces osteopenia in rats. In maturing rats, this is primarily a function of reduced formation, rather than increased resorption. Insulin-like growth factor-I (IGF-I) stimulates bone formation by increasing collagen synthesis by osteoblasts. The ability of IGF-I to prevent osteopenia otherwise cause...
